NGS (Next-Generation Sequencing) Precision Controls
Next-generation sequencing (NGS) is a technique that allows broad investigation of the genome, with high throughput, scalability and speed. However, one of the biggest challenges with this method is the number of technical errors that can impact the resulting data, from sampling through to analysis.
NGS data interpretation is challenging and prone to workflow errors that are introduced during sample preparation, sequencing, and analysis. These errors can only be partially mitigated by the use of external controls.
External controls come with their own set of challenges including:
- Inability to detect lower target-specific limit of detection (LOD) for each variant and each nucleotide position in each sample, e.g., they don't provide target-specific true LOD
- Inability to detect false positives and false negatives
- Potentially missing clinically actionable calls that are in the noise of the assay
